How quickly do the lenses transition?

Photochromic lenses respond to UV light within seconds: they begin darkening as soon as you step outside and lighten again when you come back indoors. Full transition typically takes a minute or two each way.

Will they darken in my car?

Most car windscreens filter UV, which is what triggers the lenses to darken. Photochromic glasses generally don't darken fully inside a vehicle. For driving sunglasses, you'd want a dedicated polarised pair.

Can I use these as my evening blue-blockers too?
